I have 28,000 miles on my car and I noticed that my O/D Off light cane on.  I also feel like my RPM's are running higher (0ver 4000 RPM's when driving 65 on the freeway).

Yes, this would cause the higher RPMS at highway speeds because O/D causes the vehicle not to shift as often in highway driving.
