Do I check the transmission fluid level in my Chrysler 300M when the car is off or on? Hot or cold?

I wanted to first check the level of my transmission fluid....but want to be sure I'm doing
it correctly.  My transmission is acting up - doesn't want to go from 2nd to 3rd on
occassion.  Other times...it shifts beautifully.  My car is has an automatic transmission
(2004 Chrysler 300M).  I absolutely LOVE this car!!!  Want to keep it...but I can't afford
a new transmission right now.  Anyone have any previous experience with their 300's
transmission that might be able to offer some advice?

Transmission fluid you only check with motor on Engine oil you check with motor off
