Since the transmission and the differential are bolted together at the rear of the car it could be several things making the noise. If it were mine I would first check the differential lube of metal shavings and then move on to the transmission. Does the car have a manual or automatic transmission? How many miles on the car? IF there ends up being a problem with the differential, repair of the unit is not something for the "do it yourselfer" as special tools and expertise are required to service it.

Bob has the right answer. Put it on a hoist that allows the car to operate and pin point the noise. Also check for fluid/differential oil levels. Use a magnate to see if there are metal filings in the oil. I very well could be a bearing, u-joint or drive-rod. Wheel bearing. That's why you need to locate the noise.
