Must the intake manifold or heads be removed to replace the hydraulic lifters?

Lifter intermittently ticking - at cold start, and after highway driving. Can they be replaced via valve cover removal only?

Thanks. Running some MMO & fresh oil right now (only about 160 mi., so far). Still have a tick at start-up, and ticking sets in after cruising above 2500 rpm for any length of time. This morning noticed some tick setting in idling at stop light for first time. Am wondering if lifter is being starved on feed side, or is just worn and experiencing bypass leakage. To make sure that I am understanding correctly, the V6 does not require intake manifold removal? Thanks again.
